چکیده

Ointments containing paromomycin (PA) have been used for topical treatment of cutaneous leishmaniasis. Although influence of the vehicle on skin permeation is crucial for optimization of formulations containing PA, this aspect has not been investigated experimentally. In this study, we have evaluated the influence of the formulation type [oil-in-water (o/w), water-in-oil-in-water (w/o/w) emulsions and ointment] on in vitro release and skin permeation of PA and experiments were carried out on cellulose acetate membrane and skin biopsies from hairless mice, respectively. PA release from o/w emulsion (51.7% ± 0.9 of dose applied) was faster than that observed for w/o/w emulsion (3.0% ± 0.5). PA absorption from o/w emulsion (87.1% ± 3.9) through stripped skin was 5 to 6 times higher than that observed for w/o/w multiple emulsion (14.7% ± 0.5) and ointment (16.6% ± 0.1). PA permeation through intact (normal) skin was negligible for all tested formulations. Skin uptake from o/w emulsion (0.9% ± 0.1) was greater than that observed for w/o/w multiple emulsion (0.3% ± 0.1) and ointment (0.1% ± 0.1). These results suggest that a hydrophilic vehicle (o/w emulsion) could be an interesting alternative to improve topical delivery of PA. *Correspondence:

Gelatin Microspheres for the Controlled Release of All-trans-Retinoic Acid Topical Formulation and Drug Delivery Evaluation

A topical o/w cream containing tretinoin microspheres was prepared. Gelatin microspheres of tretinoin were prepared using coacervation method. These microspheres contained about 50% w/w tretinoin. The particle size range of microspheres was between 90-150 m m. In vitro drug release from microspheres and a cream formulation was studied. It was shown that drug release from microspheres followed H...

Modeling the Rheological Properties of Carboxymethylcellulose Stabilized O/W Emulsions Based on Sunflower Oil and Tallow Fat

The aim of this research was to assess the effect of fat type and concentration as well as the continuous phase viscosity on the rheological properties of an oil-in-water (O/W) emulsion. Thus, sunflower oil and tallow fat were used as the dispersed phase of the emulsion both at 10 and 50% (w/v) and CMC solutions (at 0.1, 0.3 and 0.5% w/v) were acted as the continuous phase. CMC solutions and em...

Evaluation of the organophosphorus hydrolase enzyme activity in creams and investigation of its stability

The main purpose of this project is investigation of the organophosphorus hydrolase (OPH) enzyme activity in water in oil (w/o) and oil in water (o/w) creams and investigation of the OPH enzyme stability in formulated creams. OPH enzyme was extracted and purified from strain flavobacterium. The w/o and o/w creams were prepared using different formulations. In order to achieve an emulsion with m...

Fabrication and in vitro evaluation of Ketotifen Fumarate-loaded PLGA nanoparticles as a sustained delivery system

Abstract Ketotifen fumarate is a non-bronchodilator anti-asthmatic drug which inhibits the effects of certain endogenous substances known to be inflammatory mediators, and thereby exerts antiallergic activity. The present study describes the formulation of a sustained release nanoparticle (NP) drug delivery system containing ketotifen, using poly (D,L lactide-co-glycolide acid) (PLGA). Biodegra...
